干旱胁迫下枸杞表皮角质层蜡质积累机制及其分子基础研究

Abstract植物能通过增加表皮角质层蜡质积累提高器官保水性,响应干旱胁迫,但主要的蜡质合成途径脱羰基途径中的关键酶--醛脱羰基酶基因及其功能没有被确证。已有研究表明ECERIFERUM1(CER1)基因编码醛脱羰基酶,催化长链醛到烷的转化,对干旱敏感,但研究材料拟南芥、水稻、玉米等均为蜡质不发达、合成能力有限的中生植物,并非研究CER1基因及其功能的典型材料。本项目拟以蜡质发达的旱生经济灌木--枸杞为材料,克隆醛脱羰基酶基因(LbCER1);用real-timequantitativePCR技术定量分析LbCER1基因在干旱胁迫下的转录丰度;用RNA干扰技术沉默LbCER1基因,分析LbCER1-RNAi植株抗旱性和地上部器官表皮蜡质的形态、总含量、各组分含量变化,解析枸杞LbCER1蛋白介导的蜡质积累响应干旱胁迫的生理机制,为作物抗旱性遗传改良和培育适宜于果实干制加工的枸杞新品种提供理论依据。【英文摘要】Plantsareabletoenhanceswaterretentionanddroughttolerancebyincreasingcuticularwaxaccumulationofaerialpart.Decarbonylationpathwayisthemainpathwayforwaxbiosynthesis.However,asthekeyenzymeinthispathway,aldehydedecarbonylasegeneanditsfunctionremainelusive.IthasbeenshownthatECERIFERUM1(CER1)gene,encodedanaldehydedecarbonylasewhichcatalyzesthechemicalreactionsontransformedlong-chainaldehydeintoalkanes,playsacriticalrolesindroughttoleranceofsomeplantsuchasArabidopsisthaliana,OryzasativaandZeamaysL.ButallthesemesophytemightnotbethebestchoicetounderstandCER1geneduetotheirweakerabilitiesforwaxbiosynthesis.Inthisproject,therefore,barbarywolfberry(LyciumbarbarumL),animportantxerophyteeconomicshrubwhichhasdevelopedcuticularwax,willbeusedtounderstandthefunctionofcuticularwaxaccumulationmediatedbyCER1indroughttolerance.TheLbCER1geneencodedthealdehydedecarbonylasewillbeclonedandcharacterized.ThetranscriptabundanceoftheLbCER1geneunderdroughttreatmentwillbequantitativelyanalyzedbyusingreal-timequantitativePCR.TheLbCER1geneinbarbarywolfberrywillbesilencedbyRNAinterference(RNAi)method.Thecuticularwaxmorphology,amount,compositionanddroughttoleranceinLbCER1-RNAilineswillbeanalysed.Resultsofthisprojectshouldprovidethetheoreticalbasisinvolvedindroughttoleranceforthegeneticimprovementofcropsinthedroughtregionsandsolvingdryingproblemofbarbarywolfberryfreshfruits.
